If this part is defective, will it cause my remote start to not operate? Also, is it located above the spare tire the same as the Denali 6.2?

It will only cause it not to start if the CEL is on.

The main issue you will see with a bad vent valve is the fuel nozzle continuously clicking off while filling the tank.

Yes, the vent valve is by the spare tire.

Purge valve is on the driver side of the intake manifold.

Charcoal canister is just forward of the rear axle, almost directly above the differential.
